Neurofiziologija

Tokom procesa razvoja novih istraživačkih pravaca u Institutu, ranih 60-tih godina prošlog veka formirano je Odeljenje za kriofiziologiju pod rukovodstvom akademika prof. dr Radoslava Anđusa. Kasnije, zahvaljujući dr Miri Pašić, profesoru Biološkog fakulteta, osnovano je Odeljenje za neurofiziologiju, čiji je osnovni cilj bio ispitivanje neurofizioloških osnova ponašanja na ćelijskom nivou.

Primenom klasičnih elektrofizioloških tehnika (current clamp i voltage clamp), saradnici Odeljenja pod rukovodstvom dr Gordane Kartelije značajno su doprineli utvrđivanju osnovnih biofizičkih karakteristika pojedinačnih neurona vinogradskog puža Helix pomatia. Ova istraživanja su bila polazna osnova za dalju analizu funkcionalnih osobina neurona pod uticajem različitih aktivnih supstanci i faktora sredine (temperatura, svetlost, magnetna polja,...) i mehanizama neuromodulacije.

Pored klasičnih elektrofizioloških istraživanja na ganglijskom kompleksu puža, poslednjih godina primenom modernih tehnika analiziraju se i biofizičke osobine nervnih ćelija kičmenjaka. Metodom nametnute voltaže na deliću membrane (patch clamp) ispituju se jonske struje na nivou cele ćelije, kao i osobine pojedinačnih jonskih kanala nervnih ćelija u kulturi.

Elektrofiziološki signali analiziraju se primenom linearnih i nelinearnih matematičkih metoda (analiza talasića, fraktalna analiza), kao i razvijenih statističkih metoda (analiza nezavisnih komponenata). Primena pomenutih metoda omogućava detaljnu analizu aktivnosti pojedinačnih neurona i populacija neurona.

Delatnost Odeljenja je proširena na ispitivanje morfo-funkcionalnih karakteristika mozga i ponašanja eksperimentalnih životinja. Ispituju se efekti različitih neuroaktivnih supstanci, magnetnih polja i procesa starenja u fiziološkim i patofiziološkim stanjima. Nervne i glijalne ćelije se analiziraju metodom imunohistohemijskog obeležavanja i prate se promene nekih biohemijskih parametara u mozgu. Ponašanje životinja u specifičnim testovima (open field, plus maze, Porsolt forced swim, object recognition, light/dark box,...) automatski se prati i analizira korišćenjem Any-maze softvera.

Akcenat se stavlja i na jasno definisane neurološke i psihijatrijske bolesti ljudi. Korišćenjem animalnih modela (hemijski izazvane konvulzije kojima se sagledava epilepsija i primena aluminijuma koji se povezuje sa etiologijom Alchajmerove bolesti) ispituju se patogenetski mehanizmi koji učestvuju u nastanku pomenutih poremećaja, kao i farmakološki aktivne materije kojima bi moglo da se deluje protektivno. Postavljeni ciljevi se prate na osnovu biohemijskih i imunohistohemijskih promena u pojedinim strukturama mozga i kliničkog ispoljavanja nastalih poremećaja.

Neurobiologija

Odeljenje za neurobiologiju formirano je u okviru Instituta za biološka istraživanja "Siniša Stanković", Univerziteta u Beogradu ranih šezdesetih godina na ideju i uz angažovanje akademika Ljubisava Rakića. Tokom poslednjih 50 godina istraživanja na Odeljenju dala su značajan doprinos ispitivanju osnovnih procesa u nervnom sistemu. Odeljenje za Neurobiologiju čine laboratorije za: Molekularnu neurobiologiju, Neurohemiju i Neurofiziologiju koje ujedno predstavljaju i naučnu bazu za osnovne i doktorske studije na Biološkom fakultetu.

Odeljenje broji 40 saradnika. Članovi Odeljenja su angažovani kao predavači na osnovnim i Doktorskim studijama na Biološkom Fakultetu (BF), Univerziteta u Beogradu, na studijskim programima Molekularna Biologija i Neuronauke.

Tokom 2020. i 2021. godine saradnici Odeljenja za neurobiologiju Instituta za biološka istraživanja „Siniša Stanković" - Instituta od nacionalnog značaja za Republiku Srbiju (Laboratorija za molekularnu neuro-onkologiju) objavili su 5 publikacija u međunarodnom časopisu izuzetnih vrednosti (M21a) Drug Resistance Updates (IF = 18.5). 11. forumu Federacije evropskih društava za neuronauke u Berlinu

Saradnici Odeljenja za neurobiologiju IBISS, na 11-om forumu Federacije evropskih društava za neuronauke (Federation of European Neuroscience Societies-FENS) u Berlinu, jul 2018. godine. Naši saradnici su uživali u raznovrsnom programu najvećeg evropskog kongresa iz oblasti neuronauka i imali priliku da predstave svoja istraživanja.

Beograd će sledeće godine biti domaćin regionalnog kongresa pod pokroviteljstvom FENS, u organizaciji srpskog, turskog i rumunskog društva za neuronauke. Projekat Centra za promociju nauke (2017-2018): Biomonitoring površinskih voda

Voda je od neprocenjive važnosti za opstanak života na planeti zato zahteva posebnu brigu. Biomonitoring rečnih tokova u lokalnim zajednicama omogućava bolјu kontrolu zagađenja i jačanje pokreta za očuvanje prirodnih resursa. Allium anafazno-telofazni test se koristi za procenu toksičnih i genotoksičnih efekata uzoraka voda ili hemikalija. Primenićemo ga da ispitamo efekte koje industrijska postrojenja i komunalne vode grada mogu imati na kvalitet rečne vode. Testiranje ćemo ponoviti u dva navrata, kako bismo utvrdili i efekte vodostaja na zagađenje reka. Pored procene stanja reka cilј nam je da obučimo učenike osnovnih i srednjih škola, kao i njihove nastavnike i profesore kako da samostalno sprovode ovaj tip testiranja. Na ovaj način želimo da doprinesemo jačanju ekološke svesti kod dece školskog uzrasta i pobolјšanju kvaliteta praktične nastave u školama.

Na XIV Evropskom simpozijum za istraživanje slepih miševa (1 - 5. 8. 2017. u San Sebastijan, Baskija, Španija) Ivana Budinski dobila je nagradu za najbolju studentsku poster prezentaciju od kompanije Wildlife Acoustics. Pored diplome Ivana je dobila uređaj Echo Meter Touch Pro 2 i licencu za softver Kaleidoscope Pro.

Genetička istaživanja

Glavni i višegodišnji predmet naših istraživanja je polimorfizam B hromozoma kod žutogrlog miša, Apodemus flavicollis. U cilju rasvetljavanja biološkog smisla ovih dodatnih hromozoma u istraživanjima kombinujemo različite pristupe kao što su analiza hromozoma, geometrijska i tradicionalna morfometrija i brojni molekularni markeri (mikrosateliti, mtDNK, AFLP, RFLP). Pokušavamo da pratimo promene u genetičko-fenotipskoj strukturi u prostoru i vremenu u korelaciji sa karakteristikama životne istorije i demografskom strukturom. Cilj je da objasnimo kako se vrsta nosi sa različitim unutrašnjim i spoljašnjim pritiscima putem lokalnih adaptacija. Preliminarni rezultati koji korelišu B hromozome sa prisustvom endoparazita obećavaju u budućnosti produbljivanje ovih istraživanja. Očekujemo i značajne pomake u rasvetljavanju strukture B hromozoma kod ove vrste koji će teoriju o poreklu B hromozoma podkrepiti konkretnim dokazima.

Sem toga, bavimo se filogenijom i genetičkim diverzitetom različitih vrsta glodara, slepih miševa i zveri. U kontekstu genetičke strukture, kod ispitivanih vrsta prate se prevalenca i intenenzitet dominantnih makro i nekih mikro (virusi) parazita.

Efekti sredinskog zagađenja u vodenim i kopnenim sredinama su takođe predmet našeg interesovanja. Primenjujemo različite testove, koji mere nivoe genotoksičnosti kao što su Allium anafazno-telofazni test, mikronukleus test (na ribama) i in vivo test indukovanih hromozomskih aberacija kod sisara.
